SOX-9, along with SOX-8 and SOX-10 share at least 70% identity and comprise the SOXE group. SOX-9 is required for development, differentiation, and lineage commitment in various tissues. During embryogenesis, SOX-9 is expressed in the cartilage, neural crest, kidney, and pancreas. SOX-9 is crucial for chondrocyte differentiation and regulates transcription of the anti-Müllerian hormone (AMH) gene, as mutations in SOX-9 causes campomelic dysplasia and autosomal sex reversal. SOX-9 also exhibits several pro-oncogenic properties, including the ability to promote proliferation, inhibit senescence, and collaborate with other oncogenes in neoplastic transformation.
