It controls the expression of genes involved in pancreas specification, morphogenesis, and beta cell differentiation. PDX1 is essential for the formation of pancreatic buds from the foregut endoderm and the subsequent differentiation of endocrine, exocrine, and ductal cell lineages within the pancreas. In the adult pancreas, PDX1 is predominantly expressed in beta cells, where it regulates the expression of genes involved in insulin biosynthesis, secretion, and glucose sensing. PDX1 acts as a transcriptional activator of the insulin gene (INS) and other genes required for beta cell function and survival. It coordinates the expression of multiple genes involved in glucose metabolism and maintains beta cell identity and function. Mutations in the PDX1 gene or alterations in its expression levels have been associated with both type 1 and type 2 diabetes mellitus.
