ZytoLight products are designed for the identification of genetic aberrations e.g. translocations, deletions, amplifications, and chromosomal aneuploidies by fluorescence in situ hybridization (FISH) in formalin-fixed, paraffin-embedded tissue sections, cell samples, blood or bone marrow smears, and metaphase chromosome spreads.

Detection Method
ZytoLight FISH

The ZytoLight system uses direct labeled FISH probes, eliminating the need for secondary fluorophore-coupled antibody detection.
An excitation filter restricts the full-spectrum light emitted by the microscope lamp to a defined wavelength that specifically excites the fluorophore attached to the probe. A dichroic mirror then directs this light onto the specimen. The fluorophore emits light of longer wavelengths that passes the mirror. Finally, a barrier filter reduces the emitted light to a detectable defined wavelength.

ZytoLight FISH probes are directly labeled using the ZytoLight Direct Label System II to support strong signal intensity and clear visualization. ZytoLight single-copy (SPEC) probes incorporate the ZytoLight Repeat Subtraction Technique, reducing non-specific background and eliminating the need for additional blocking of repetitive sequences. ZytoLight CEN probes target repetitive human satellite DNA sequences within chromosome centromeric regions, producing bright, chromosome-specific signals.

Convenient Kit Solutions

ZytoLight FISH probes can be used with the ZytoLight FISH-Tissue Implementation Kit to simplify FISH workflows on tissue specimens. The kit includes pretreatment solutions, wash buffers, DAPI/Anti-fade solution and a detailed protocol to support consistent implementation. For select targets, complete kits are also available that include both the probe and all required reagents for FISH analysis.
